Comparison of efficacy between three clinical samples (blood,urine, semen) for molecular detection of Brucella Spp infection in maledogs

Background and Aim : Dogs are the main hosts of Brucella canis. Infection with B. canis isconsidered zoonosis disease, although it rarely seen in humans. Other species that cause brucellosisin dogs include B. abortus, B. melitensis and B. suis. The symptoms are mainly genital andabortion is common in ۷ to ۹ weeks of pregnancy. Embryonic absorption has also been reported ۲to ۳ weeks after breeding. The normal morphology of the sperm is altered in infected males.Sometimes inflammation of the epididymis, testes and scrotal edema is occurred. Male dogs oftenshow irreversible infertility despite treatment. Studies have indicated the inability to clear theinfection of the prostate gland, so the identification and removal of infected dogs from the kennelhave particular importance.Methods : In this study, ۳۷ male dogs in kennels with reduced reproduction were selected. Aftercomplete clinical examinations, blood samples were taken from cephalic vein. Semen and urinesamples were taken by digital manipulation and catheterization method. DNA was extracted byGeneAll® ExgeneTM extraction kit by manufactured instruction. Brucella spp. was confirmed byPCR with specific primers IS۷۱۱ (Ouahrani-Bettache ۱۹۹۶).Results : From ۳۷ breeding dogs, ۱۴(۴۳.۷۵%) of urine,۱۱(۲۹.۲۷%) of semen and ۴(۱۰.۸%)samples were positive, which represents very significant infection rate. There was a significantrelationship between history of contact with farm animals and positive PCR test results (P = ۰.۰۴۵).In comparison with blood (۱۰.۸%) and semen (۲۹.۷۲%), urine is a very good clinical sample forevaluation of brucella infection in male dogs.Conclusion : Due to the high prevalence of Brucella infection in breeding male dogs in kennelsand considering the fact that each male dog mates with at least ۲۰ to ۳۰ female dogs, male dogsmonitoring before entering the kennels for brucellosis is highly recommended. The zoonoticaspects and economical loss of disease are both very crucial so definitive diagnosis is veryimportant and the selection of the best clinical samples and diagnostic methods are very essentialwhich is noted in the present investigation. our study provides evidence of the usefulness of theurine and semen samples for the diagnosis of canine brucellosis as much as that of blood samples.
