Analysis of MicroRNA-۱۴۶a gene polymorphism in patientswith brucellosis: A Case-Control Study

چکیده
Background and Aim : Single nucleotide polymorphisms (SNPs) represent the most abundantgenetic variation in the entire human genome. MicroRNAs (miRNAs) are a type of evolutionaryconserved small single-stranded non-coding RNAs which control the target gene expressionnegatively via mRNA degradation or translational expression at the post-transcriptional level.MiR-۱۴۶a was first recognized as a regulator of the immune system responding to microbialinfections. The aim of the present study was to investigate the association between the risk ofbrucellosis and genetic variations in miR-۱۴۶a.Methods : In this case-control study included ۱۰۸ Brucellosis patients and ۱۰۸ healthy controls.We genotyped two SNPs (rs۲۹۱۰۱۶۴ and rs۵۷۰۹۵۳۲۹) of the miR-۱۴۶a using tetra-primeramplification refractory mutation system-polymerase chain reaction (T-ARMS-PCR) andrestriction fragment length polymorphism-polymerase chain reaction (RFLP-PCR) methods.Results : Co-dominant (P = ۰.۰۴۷) and recessive (P = ۰.۰۱۸) models were significant at positionrs۵۷۰۹۵۳۲۹ between the two groups of patient and healthy. The rs۲۹۱۰۱۶۴ SNP was significantlyassociated with brucellosis in co-dominant [OR = ۴.۲۷, ۹۵% CI = (۲.۳۵–۷.۷۹, P = ۰.۰۰۱] anddominant [OR = ۳.۵۲, ۹۵% CI = (۱.۹۷–۶.۳۰, P = ۰.۰۰۱] models. The A C haplotype (rs۲۹۱۰۱۶۴and rs۵۷۰۹۵۳۲۹) was associated with brucellosis in the assessed population [OR (۹۵% CI) = ۱.۹۸(۱.۲۲–۳.۲۰), P = ۰.۰۰۵۹].Conclusion : Our study showed significant differences in genotype and haplotype frequencies ofmiR-۱۴۶a variants between brucellosis and controls. Further studies on the larger sample sizes arerequired to verify the observed associations.کلیدواژه ها
