Investigation of the relationship between the gut microbiotaand inflammatory bowel disease in a mouse model

چکیده

Background and Aim : Aim: Inflammatory bowel disease (IBD) is a group of chronicgastrointestinal disorders affecting millions of people worldwide. Several factors are involved inthe development of this disease, but the gut microbiota is known to be one of the most importantfactors. The aim of this study was to determine the relationship between gut microbiota and IBDusing mouse model.Methods : Methods: In this study, two methods were used: chemical induction with dextran sulfatesodium (DSS) and biological induction with stool from human with IBD (fecal microbiotatransplantation) to induce inflammation in the gut of mice. The populations of the gut microbiotain both groups were studied using real-time PCR. In addition, the serum levels of inflammatorycytokines and the colon tissues of the mice were analyzed.Results : Results: The pathological results showed that the colon tissue in the FMT group hadinflammatory changes as in the DSS group. The changes in the intestinal microbiota population inboth FMT and DSS groups on the last day of the study also showed a similar pattern, such that thebacteria belonging to Phyla Actinobacteria, Firmicutes and Bacteroidetes decreased, while thepopulation of γ-Proteobacteria increased significantly. An increase in serum levels of IL-۱ and IL-۶ was observed in both groups.Conclusion : Conclusion: The results of this study show that there is a mutual relationship betweenthe gut microbiota and inflammatory diseases, and that the gut microbiota is not only the cause ofIBD but may also be a consequence of this disease. In fact, by chemically inducing inflammation,the gut microbiota was altered. On the other hand, performing FMT from human stool with IBDaltered the gut microbiota of mice and induced inflammation in the mouse model
