Background and Aim : Glycopeptides such as vancomycin are frequently choice antibiotics forthe treatment of infections caused by methicillin resistant Staphylococcus aureus (MRSA). For thelast years incidence of vancomycin intermediate S. aureus (VISA) and vancomycin resistant S.aureus (VRSA) has been increasing in various parts of the world. The present study was carriedout to find the presence of VISA and VRSA in Thi-Qar province-south of Iraq.Methods : A total ۳۱۵ staphylococcal isolates consisting of ۱۳۰ S. aureus and ۱۸۵ coagulasenegative staphylococci (CoNS) were isolated from different clinical specimens from varioushospitals including AL Hussein teaching hospital, AL Haboubi teaching hospital, AL Musawihospital for children, and Burn center. All S. aureus isolates were subjected to MIC testing (againstvancomycin) on Brain Heart Infusion (BHI) vancomycin screen agar test. Disc diffusion testingfor Ampcillin (۱۰ ug), Oxacillin (۱۰ ug), Gentamicin (۱۰ ug), Amikacin (۳۰ ug) Vancomycin (۳۰ug), and cefoxitin (۳۰ ug) was performed. The presence of mecA, vanA, vanB, vanC, vanD, and۱۶s rRNA genes was evaluated by PCR.Results : Out of ۱۳۰ S. aureus, ۷ strains were found to be vancomycin resistant. Seven strains hadgrowth on BHI vancomycin screen agar (vancomycin ۶ ug/ml). All isolates were positive for mecAgene. vanA, vanB, and vanC were detected in ۳, ۲, and ۲ isolates, respectively. vanD gene was notdetected.Conclusion : Conclusion: The present study for the first time reveals the emergence of VRSAfrom this part of south Iraq and indicates the magnitude of antibiotic resistance in and around thestudy area. The major cause of this may be unawareness and indiscriminate use of broad-spectrumantibiotics.
