The application of gene expression analysis by ART in the selection of competent genes for pregnancy prediction

Backgrounds: In human assisted reproductive techniques (ART), selecting high-qualityembryos for transferring is usually based on morphological criteria. Analyzing gene expressionof Cumulus cells might lead to important molecular information about the embryo quality.CALM۱. One of the well-known causes of infertility is polycystic ovary syndrome (PCOs). Thenumber of retrieved oocytes with high implantation potential in PCOs patients is limited;therefore the process of selecting a good embryo in PCOs patients receiving ART is veryimportant.Materials and Methods: Granulosa cells were collected from ۳۳ infertile patients with PCOs asan experimental group and ۳۳ patients who referred to an infertility center for egg donation as acontrol group.Results: Expression of all three genes CALM۱, PSMD۶, and AK۱۲۴۷۴۲ in the pregnant groupwas higher than the non-pregnant group. Although the increase was not significant for theCALM۱ gene, it was significant for the other two genes: PSMD۶ (p <۰.۰۰۱) and AK۱۲۴۷۴۲ (p<۰.۰۵). The expression of CALM۱ and AK۱۲۴۷۴۲ genes increased significantly and theexpression of PSMD۶ decreased significantly in PCOs group compared to the control group (p<۰.۰۵).Conclusion: If our prediction model is based on differences in gene expression levels betweenpregnant and non-pregnant groups then all three genes will be proper markers for predictingembryo competence due to increased expression levels in pregnant groups. If our predictionmodel is based on comparison of gene expression levels between control and PCOs groups, thesethree genes cannot be introduced as proper markers for predicting embryo competence due toineffectiveness of gene changes on fertility outcomes in PCOs group.
