The effect of Composol Medium on platelet microRNAs expression during blood bank storage

Backgrounds: Platelet additive solutions, such as Composol-PS, are good choices for extendingthe storage time of platelet concentrates (PCs) in blood banks. Platelets express high levels ofvarious microRNAs (miRs). We aimed to investigate platelet miR-۱۶, miR-۱۲۷ and miR-۳۲۰aexpression during storage and the effect of Composol medium on miRs expression.Materials and Methods: Ten PC bags from healthy volunteers were collected and each bag wasdivided equally into two separate bags, with or without Composol, bags were stored for sevendays and tested for platelet functional tests and miRs expression analysis. Statistical analysis andmiRs expression interpretation were fulfilled by SPSS and REST. A P<۰.۰۵ was consideredstatistically significant.Results: All miRs expression was elevated during the storage in both plasma and Composolmedium, but only the results of miR-۱۶ were statistically significant (p<۰.۰۵). The most obviousup-regulation were reported for day ۵ of storage in both plasma (mean fold change = ۵.۰۲, p =۰.۰۰۲) and Composol (mean fold change = ۵.۴۲, p = ۰.۰۲۶) groups. Furthermore, PCs stored inComposol medium showed better results according to the platelet functional tests and miRsexpression. Despite the overt increased expression of miR-۱۶ in both groups, its expressionsignificantly decreased in the Composol samples (mean fold change =۰.۳۳۹) compared with theplasma samples (p<۰.۰۵). Results for miR-۱۲۷ and miR-۳۲۰a were less obvious and notsignificant.Conclusion: We concluded that Composol might be a good additive solution for extendingplatelet storage time, compared with conventional plasma environment. This may be as a resultof miR-۱۶ down-regulation in Composol medium.
