Exploration of potential key genes in oral squamous cell carcinoma by gene network analysis

Backgrounds: Oral cancer is one of the most prevalent malignancies in the world, with adelayed clinical diagnosis, poor prognosis and costly treatment options. Oral cancer is a kind ofmalignant neoplasia that develops on the lip or oral cavity. It is usually classified as an oralsquamous cell carcinoma (OSCC) since squamous cells are histologically responsible for ۹۰% ofmalignancies in the dental field. Oral cancer is a preventable illness in which smoking andalcohol—both significant risk factors—are present in ۹۰% of cases, both having a synergisticimpact. Patients are managed using traditional histologic criteria such as TNM and tumorgrading, although novel molecular and cellular markers have been explored to enhance therapyand survival.Materials and Methods: Microarray dataset GSE۳۰۷۸۴ was downloaded from gene expressionomnibus (GEO). Then, the transcriptome analysis console (TAC) was used to normalized andanalyze differential expression genes (DEGs). The genes with adjusted p-value (FDR)< ۰.۰۵ and|𝑙𝑜𝑔۲𝐹𝐶| > ۱.۵ were selected as DEGs between normal and OSCC samples. String, cytoscapeand gephi were used to construct protein-protein interaction (PPI) and visualization, respectively.Results: ۲۶۰۱ genes were obtained as DEGs (۱۳۳۵ upregulate, ۱۲۶۶ downregulate). Our analysisrevealed ۵ hub genes, namely, IL۶ (interleukin ۶), MYC (v-myc avian myelocytomatosis viraloncogene homolog), ITGB۱ (integrin beta ۱), PTPRC (protein tyrosine phosphatase, receptortype, C) and MMP۹ (matrix metallopeptidase ۹). Furthermore, the results of the KEGG pathwayanalysis revealed that these genes were enriched in significant pathways, including proteoglycansin cancer, focal adhesion and pathways in cancer.Conclusion: Consequently, the critical genes and pathways associated with the pathogenesis andprognosis of OSCC were identified by bioinformatics analysis. Studying the regulatory networkwould lead us to create a competent approach to prediction models of the illness.
