Backgrounds: It has been shown that curcumin has effective anti-cancer properties. However,the absorption efficacy of curcumin is too low to have notable achievement in therapy. Toovercome this problem, we employed Gemini Surfactant nanoparticles to improve the stability ofcurcumin, and then studied the metastatic effects of this compound on the AGS gastric cancercell line.Materials and Methods: Human gastric carcinoma AGS cell lines were treated with Geminicurcumin, and cell viability was assessed by MTT assay. The scratch test was performed toevaluate the metastasis of cancer cells. The expression of the noncoding RNA CCAT۲ gene andits downstream c-MYC was evaluated by qPCR and western blotting.Results: Gemini curcumin affected the cell viability of AGS cells in a dose- and time-dependentmanner with IC۵۰ values of ۵۷.۹۲ μM and ۳۸.۸۵ μM at ۲۴ h and ۴۸ h, respectively. Our datashowed that Gemini curcumin nano compound down-regulates lnc-CCAT۲ and c-MYC genes.Scratch test also confirmed its anti-metastatic properties on AGS cells.Conclusion: Taken together, our results confirmed anticancer effect of Gemini curcumin ongastric cancer. However, further studies on molecular level are needed to demonstrate thisfinding.
