چکیده

Backgrounds: Acute lymphocytic leukemia (ALL) is a type of cancer of the blood. In theUnited States, the majority of ALL cases occur in children, with an incidence of ۳ per ۴/۱۰۰,۰۰۰in patients aged ۵ to ۱۴. The purpose of this research was to determine how Nithiosemicarbazonecomplexes altered the expression of the NKILA in an acute lymphoblasticleukemia cell line.Materials and Methods: Two concentrations of Ni Thiosemicarbazone complexes wereprepared: ۱۰۴μM at ۲۴h and ۵۱μM at ۴۸h. The JurkatE۶.۱ cell line was purchased from PasteurInstitute and treated with prepared doses of the Thiosemicarbazones complexes Ni at ۲۴ and ۴۸hafter cell passage. The expression changes of NKILA and GAPDH were studied using Real-Time PCR after RNA extraction and cDNA synthesis. Finally, Rest ۲۰۰۲ Software was used toanalyze the data.Results: The results showed that the expression of NKILA in comparison with the GAPDHdecreased after ۲۴h of Ni treatment at ۱۰۴μM. According to the findings, changes in NKILAgene expression increased after ۴۸h at ۵۱μM that increase was statistically significant Thesechanges included ۱۰۴μM (۰.۸۹۶) at ۲۴h and ۵۱μM (۱.۲۱۴) at ۴۸h, respectively (P < ۰.۰۰۱).Conclusion: According to the present study results, alternation in NKILA expression aftertreatment with Ni, at ۵۱μM was effective in increase of LncRNA NKILA expression. Evidenceshowed that the Ni requires a longer time and lower concentration for better efficacy because thedrug was ineffective in increasing gene expression at higher concentrations in ۲۴h.
