Backgrounds: Cancer cells continue to reproduce, and for this to happen, their telomerase mustbe very active. This is not the case with healthy cells. To reduce telomerase activity, we need toknow how to target the activity of this enzyme. Telomerase is an essential enzyme for sequentialcell proliferation in metastatic cells. TRBF proteins are based on binding sites, on the telomericregions, as well as the presence of these proteins increases and decreases telomerase activity.Materials and Methods: The aim, was to collect and compare TRBF gene family sequences tofind conserved motifs and to investigate the affinities of this gene, it was different betweenmammalian species. Using RT-PCR, the coding sequence of a TRBF peptide was isolated andidentified. For similarity studies with the help of Mega bioinformatics Software, the obtainedsequence was compared with the same TRBF sequence in other mammalian species.Results: In this work, for the first time, studies have been carried out on ۳ levels of protein,mRNA and, DNA. As the sequence of telomeric region sequences of organisms is constantlyincreasing and decreasing, information of these regulatory proteins and synthesis of the abovetelomeric region in different mammalian species has been considered. TRBF affinities betweendifferent Mammalian species were reported in the N-terminal and C-terminal segments.Conclusion: Specific conserved amino acid fragments and the second reverse transcriptase wereintroduced as the functionally conserved motif of all species at the protein level. Finally, directinhibition of telomerase and immunotherapy of telomerase can be a method of inhibiting cancers
