Evaluation of miR-۳۸۲ expression change in the prefrontal cortex of the brain following alcoholism in male Wistar rats

Backgrounds: Alcohol has various side effects and its abuse causes severe metabolic andphysiological effects in all body systems. Given that different area of the brain, including theprefrontal cortex, play a role in causing addiction; this study aimed to investigate the geneticexpression of miR-۳۸۲ in the prefrontal cortex of the brain in rats.Materials and Methods: Fifty male Wistar rats weighing ۲۰۰ to ۲۵۰ g were prepared. Adultmice were kept under hygienic conditions at room temperature with free access to water andfood for ۱۲ hours in a light /dark period. Alcohol was administered to animals orally for ۲۱ daysat a dose of ۵ mg/kg. The mouse's head was removed and then the skull was split and theprefrontal cortex was removed. The expression of miR-۳۸۲ relative to the reference gene wasexamined using real-time PCR in this region. Statistical analysis was performed using SPSSsoftware.Results: The results showed that following the consumption of alcohol for ۲۱ days, theexpression level of miR-۳۸۲ in the prefrontal cortex significantly decreased (P <۰.۰۵).Conclusion: The study of miRNAs as mediators of molecular mechanisms that also affect theexpression of other genes can increase our understanding of the mechanism of complications ofproblems such as alcoholism.
