Backgrounds: Breast cancer is the major cause of death from cancer among women. Given thedrug resistance created in the treatment of this disease, it is very important to identify newtherapies and new anti-cancer drugs. Some studies indicate the cytotoxic effects of vanadyl oxidesulfate (VOSO۴). Therefore, this study aims to evaluate the anti-cancer effect of VOSO۴ in thetreatment of breast cancer.Materials and Methods: MCF-۷ cell line was treated with different concentrations of VOSO۴for ۲۴ and ۴۸ hours. The measurement of cell death was performed by MTT assay. The cellapoptosis rate was measured using Annexin V / Propidium Iodide Assay through flow cytometry.The level of expression of p۵۳, P۲۱, Caspase۸, Sod۱, Sod۲, and Bcl۲ mRNAs were evaluated,and Western blotting was performed for Sod۱ to confirm the results.Results: The results showed that the half-maximal inhibitory concentration (IC۵۰) for VOSO۴was ۲۵ and ۲۰ for ۲۴ and ۴۸ h, respectively. Indeed, VOSO۴ has dose-dependent cytotoxiceffects on the MCF-۷. Also, the apoptosis of the cells after ۲۴ h of exposure to VOSO۴ was morethan ۵۲%. Moreover, after ۲۴ hours of exposure to VOSO۴ with IC۵۰ concentration, theexpression of p۵۳, P۲۱, Caspase۸, Sod۱, Sod۲ mRNAs increased and the expression of Bcl۲mRNA decreased. The Western blotting on Sod۱ confirmed these findings.Conclusion: Our results demonstrated that VOSO۴ has an apoptotic and cytotoxic effect onbreast cancer cells. Therefore, it is likely that this substance can be considered as acomplementary agent for the medical treatment of breast cancer patients.
