Naringenin and Resveratrol Anti-tumor Impact on the Y۷۹ Retinoblastoma by Affecting E/N-Cadherin and Galectin-۳; PossibleSynergistic Effect

چکیده
Backgrounds: One of the detrimental features of retinoblastoma is highly invasiveness of thistype of cancer; the ability to metastasize into distal organs even in the early stages. Thisphenomenon highlights the importance of experiments targeting this characteristic to diminishthe disquieting outcomes. In this study, our main aim was to assess the impact of naringeninand/or resveratrol treatment on the important genes expression in the metastasis and cancerprogression pathways in the Y۷۹ retinoblastoma cell line.Materials and Methods: To attain the cytotoxicity dose of both reagents, an MTT assay wasperformed for ۲۴ and ۴۸ hours. The Y۷۹ cells were then treated with a lower dose comparedwith the IC۵۰. To further investigate the synergistic effect of the compounds, concurrenttreatment was also done. Finally, the expression of E-cadherin, N-Cadherin, and Galectin-۳ wasinvestigated in different samples with the aid of real-time PCR.Results: According to the MTT assay the ۲۴ hours IC۵۰ for resveratrol was about ۱۰۰ μg/ml and۴۸ hours IC۵۰ was about ۵۰ μg/ml. Naringenin ۴۸ hours IC۵۰ was calculated to be ۱۰۰ μg/ml.Treatment of Y۷۹ cells with naringenin, resveratrol, or the concurrent treatment down-regulatedthe N-Cadherin mRNA expression level. Treatment with resveratrol or the concurrent increasedthe expression level of the E-Cadherin and diminished Galectin-۳ expression.Conclusion: Resveratrol seems to be more toxic for Y۷۹ cells compared with naringenin. Twocompounds didn’t show a significant synergistic effect. Both compounds exert an anti-metastaticeffect on these cells by inducing N-Cadherin down-regulation. Resveratrol enhanced theexpression of the E-Cadherin, along with the decreasing the Galectin-۳ exerts even more antitumoractivity, and can be proposed as a beneficial reagent in cancer immunotherapy.کلیدواژه ها
