چکیده

T-cell acute lymphoblastic leukemia (T-ALL) is a high-risk hematologicalmalignancy that affects early-developing T-cells, accounting for ۱۵% ofALL cases in children. Studies in this subject are attempting to uncovergenes involved in lymphocyte differentiation and death, as well as essentialcellular pathways such as ERK/MAPK, which could aid in a betterunderstanding of leukemia pathogenesis. This is a review that wasconducted by searching Google Scholar, PubMed, and science direct aboutERK/MAPK signaling pathway function in acute lymphoblastic leukemia.As a result, the MAPK signaling pathway is involved in a variety ofphysiological activities in cells, including cell growth, development,division, and death. The MAPK family includes ERK, and the ERK/MAPKsignaling pathway is at the heart of the signaling network that controls cellgrowth, development, and division. The MAPK tertiary enzymatic cascade,which includes an upstream activator sequence, MAP۳K, MAP۲K, andMAPK, is followed by the basic signal transmission steps. Ras is anupstream activating protein in the ERK pathway, Raf is MAP۳K,MAPK/ERK kinase (MEK) is MAPKK, and ERK is the MAPK, constitutingthe Ras-Raf-MEK-ERK pathway. In conclusion, MAPK-ERK pathwayinhibitors, particularly MEK inhibitors, impede the inactivation of proapoptoticin T cells, resulting in cell death. The synergistic action of MEKinhibitors is not confined to IL۷-induced signaling or the presence ofparticular activating (mutational) events, highlighting the relevance ofMAPK-ERK signaling in T-ALL.
