Therapeutic effect of recombinant adenovirus on AGS cancer cell line

چکیده

Oncolytic viruses are tumor selective and potential anticancer agents. Adenovirus type ۵ is the most commonly used oncolytic agent for treating tumors. The poor expression of the viral receptor on the surface of tumor cells is a major problem in the use of adenovirus type ۵. The study aimed to improve the attachment of the modified adenovirus to tumor cells by applying an analog of somatostatin [SST] [i.e., octreotate]. In the adenovirus fiber knob's HI-loop, the octreotate sequence was introduced. By utilizing the calcium phosphate technique, modified virus was transmitted to HEK ۲۹۳ packing cells, and a new virus was generated. Using the TCID۵۰ technique, the virus titer was determined. The human adenocarcinoma stomach cell line was challenged with modified adenovirus and wild-type ۵ strain [AGS]. Next, compounds were investigated for cytopathic impacts. The modified virus was validated employing PCR and sequencing. In comparison to the wild-type adenovirus, the modified adenovirus was found to be more capable of infecting AGS cells. Experiments showed that over ۸۱% of AGS cells treated with fiber-modified Ad۵ were killed after three to four days. This study suggests that fiber-modified Ad۵ is an oncolytic agent against AGS cells.
