Evaluating the effects of a liposomal formulation containing an oligosaccharide isolated from Rosa canina on the sensory-motor function in a rat model of spinal cord injury

Background and Objective Spinal cord injury (SCI) is one of the most important causes of sensory-motor disorder. Following SCI, the spinal cord passes through the primary and secondary phase which causes a permanent neurological deficit. Due to inflammatory, oxidative stress and apoptotic properties in the secondary phase of SCI, and the anti-inflammatory, antioxidant and anti-apoptotic effects of oligosaccharides on the other disorders of nervous system and the sustained release manner of liposomal-oligosaccharide can alleviate the hallmarks of SCI.Materials and Methods The liposomal formulation of oligosaccharide was prepared using thin film hydration method. The fabricated liposomes were evaluated in terms of properties such as size, zeta potential, shape and loading efficiency of the drug by using DLS, SEM and HPLC, respectively. In general, wistar male rats were divided into five groups of Sham, SCI and Nano-oligosaccharide (۰.۶, ۱.۲, and ۱.۸ mg/kg). Aneurysm clip was used to induce injury in the SCI and Nano-oligosaccharide groups. Treatment with nano-oligosaccharide or the vehicle (empty liposome) was carried out intrathecally. The sensory-motor function and weight changes evaluated once a week for four weeks.Findings The optimum size of nano-oligosaccharide was ۱۱۸ nm and oligosaccharide encapsulation efficiency in Nano-liposomes was ۶۰.۶۷%. The release of nano-oligosaccharide displayed sustained release behavior over the period of ۴۸h. Nano-oligosaccharide group compared with the SCI group, significantly improved motor function evaluated by inclined plain and BBB test, as well as sensory improvement evaluated hotplate and acetone drop test. Besides, Nano-oligosaccharide reduced weight loss after SCI.Conclusion Altogether, we concluded that nano-oligosaccharide is a promising therapeutic agent for medical therapies with advantageous effects to enhance motor function. Further studies are needed to confirm the health benefits as well as the cellular and molecular mechanism of Nano-oligosaccharide as a promising drug in the treatment of SCI.
