Sampling and analysis of a tree stem for biotechnology studies, specially enzymatic activity, can be further complicated by the induction of enzymatic activity associated with the tissue wounding. Mechanical punches are the tool currently used to sample tissues within the stem of a tree. These punches can remove a tissue sample from the tree, but the period of time from initial sample collection and wounding to cryofixation can be minutes; long enough to induce a variety of enzymatic reactions which can alter enzymes in the sample. Those reactions potentially cloud results and interpretation of data. A new tool, named the freezer sampler (patent pending no° PD۴۲۴۸۴۸۳۱ in US), was specifically designed for sampling tree stem tissue for enzyme analysis. Freezer sampler is designed to rapidly obtain tissue samples of different size (۴cm, ۳ cm diameter). The cutting tube-like blade is inserted into the tree stem using a rotary motor force and no other accessory is required.
