MOV۱۰L۱ and PLCL۲ genes expression in mice transgene embryonic stem cells (OCT۴-GFP) induced by retinoic acid and human follicular fluid

چکیده

The study aims to generate stem cells induced by retinoic acid and human follicular fluid to utilize them in the treatment of infertility. In this experimental study, OCT۴-GFP transgenic embryonic stem cells were analyzed in the presence of ۲۵% and ۱۵% follicular fluid concentrations alone and with retinoic acid for ۷, ۱۴, and ۲۱ days, and the expression of MOV۱۰L۱ and PLCL۲ genes has investigated. In all time, the MOV۱۰L۱ gene at both concentrations of retinoic acid had a maximum expression rate of ۳۰۱ times that of embryonic stem cells. The expression of the PLCL۲ gene has declined, provided it has risen at a concentration of ۵ μM retinoic acid, which means the positive and continuous effect of this inducer on the expression of the gene. Also, in all time, the expression of both genes in the presence of follicular fluid declined and was lower compared to the control group. This status in all three times in follicular fluid groups is ۲۵% less than the ۱۵% groups. The study revealed that mice embryonic stem cells differentiate in the presence of human follicular fluid; however, the differentiation process is not in the procedure of generating cells such as male germ cells that can express the MOV۱۰L۱ and PLCL۲ genes. However, the presence of retinoic acid has stimulated gene expression.
