A n ovel intera ction bet ween Protein Z and Z-depen dent Prote ase Inhibitor complex

Protein Z (PZ) is a multi dom ain VKD dependent protein which contains GLA, EGF۱, ۲ and SP domain. PZ has a cofact or role as m ediating in hibition of the Factor X (FX) by the Z-dependent protea se inhibitor (ZPI) in the presence of Ca۲+ ions and phos pholipids [۱]. PZ circul ates in plas ma in a com plex with ZPI. In the previous studies, interac tion betwee n PZ and ZPI has been found and involves ionic and hydr ogen bond i nteractions between Glu -۲۴۴, His-۲ ۵۰, Arg-۳۵۰, Gln-۳۵۷, His-۲۱۰, A rg-۲۹۸, Arg-۲۱۲, and G lu-۲۱۹ fro m protein Z with Lys-۲۳۹, Asp-۲۳۸, Asn-۲۶۱, Asp-۷۴, Asp-۲۹۳, Thr-۲۹۶, and Lys-۶۸ from ZPI. In the summery, they showed that P Z bind to ZP I with SP d omain (resid ues ۱۳۵-۳۶ ۰). But our results show that PZ can bind to ZPI with EGF۲ domain as w ell as SP do main [۲].
