An efficient synthesis of ۲-Acetoxy-benzoic acid cyclohexylcarbamoyl-aryl-methyl esters in water using one-pot three-component approach under ultrasonic irradiation and without catalyst has been described. This rapid method produced the products in short reaction times (۳–۵ min) and excellent yields (۸۵–۹۵%). The structures of the products were deduced from their spectroscopic data (FT-IR, ۱H NMR, ۱۳C NMR). The in vitro antibacterial activities of this compounds were evaluated against gram-positive and gram-negative bacteria using the cultures of three different standard microorganisms. Most of the compounds prepared as part of this study exhibited good antibacterial activity against Pseudomonas aeruginosa, Acinetobacter calcoaceticus and Bacillus subtilis
