Investigation of human interleukin ۶(۱۷۴) gene rs۱۸۰۰۷۹۵ polymorphism in patients with Psoriasis using ARMS-PCR technique

چکیده

Psoriasis is a chronic inflammatory skin disease that is prevalent among ethnic groups. IL-۶ is a multifunctional cytokine that is involved in host defense, immune responses and inflammatory responses. The IL۶ gene with rs۱۸۰۰۷۹۵ is located on the short arm of chromosome ۷. In the IL۶ promoter region there are four polymorphisms located at positions (G/A) -۵۹۷, (G/C) -۵۷۲, (A / G) -۳۷۳, (G/C) -۱۷۴. In this study, rs۱۸۰۰۷۹۵ was evaluated at (-۱۷۴) G/C. The aim of this study was to compare the frequency of IL-۶, rs ۱۸۰۰۷۹۵ (-۱۷۴) G/C gene polymorphism in patients with psoriasis and healthy individuals. This case-control study was performed on ۵۰ patients and ۲۳ healthy individuals in west Mazandaran and east Guilan. After extracting DNA of blood samples, (-۱۷۴) G/C IL-۶ gene polymorphisms was studied using ARMS PCR method. The results were validated using DNA sequencing method. Data analysis were performed with SPSS۲۲ software. The frequency of CC, CG and GG genotypes in the control group were ۴۴%, ۳۹% and ۱۷%, respectively, and in the patient group were ۲۲%, ۲۶% and ۵۲%, respectively. So this difference is significant at the ۰.۰۵ level (p = ۰.۰۱۸). Also the frequency of G and C alleles in the control group were ۶۳% and ۳۷% and in the patient group ۳۵% and ۶۵%, respectively, Therefore, this difference is significant at the ۰.۰۵ level (p = ۰.۰۰۲). Based on the results, it can be said that IL-۶ G/C gene polymorphism is likely to play an important role in causing psoriasis.
