چکیده

Background and Aim : Halophilic bacteria live in many extreme enviroments under high-salt stress. Identification of novel halophilic bacteria can be very important in biotechnological and ecological studies. In This study, we survey heterotrophic bacterial diversity in sediments from saline wetland in south of Halghe Dare hills as one of the hypersaline region in Alborz province.Methods : The sampling was performed from ۰-۲۵ cm depth. Isolation of heterotrophic bacteria was conducted using R۲A agar medium. After differentiation of isolates based on morphological and biochemical characteristics, identification and phylogenetic relationships analysis of selected isolates were performed by ۱۶S rRNA gene sequencing and analysis using NCBI databases and bioinformatics softwares.Results : From the ۱۰۲ purificated isolates, ۱۶ strains were selected for ۱۶S rRNA gene sequencing which included ۱۵ species into ۹ genera including Bacillus (۳۲.۲%), Halomonas ۲۵%, Virgibacillus (۶.۲%), Gracilibacillus (۶.۲%), Streptomyces (۶.۲%), Aminobacter (۶.۲%), staphylococcus (۶.۲%) , Brevibacterium (۶.۲%) and Planococcus (۶.۲%). The number of ۶ isolates due to the similarity of less than ۹۸.۷% in ۱۶S rRNA gene sequence to the standard species can present novel bacterial species.Conclusion : Presence of various types of bacterial genera and some novel taxonomic groups in saline wetland in south of Halghe Dare hills as one of the extreme ecosystem in central regions of Iran presents a new genetic and microbial source.
