Anti-cancer potential of Actinobacteria isolated from Iran

Background and Aim : Nowadays actinobacteria were known as the source of secondary metabolites with diverse biological activities and structures. These compounds can be promising sources of novel anti-cancer drugs. According to Bacterial products reduction, actinobacteria isolation from the unique ecosystems to find novel bioactive compounds is an interesting approach. This study for assessment of production of anti-cancer compounds from native Iranian actinobacteria.Methods : The samples were collected from different locations like Persian Gulf water, soil and plants root. Serial dilution was prepared (۱۰-۱ - ۱۰-۴), then the diluted samples were inoculated in media (isp۲ and isp۴) and incubated at ۳۰°C for ۱۰ days. After that, the isolated strains were cultured in a ۲۰۰ mL broth media and incubated in ۳۰°C with ۱۴۰ rpm for ۸ days. For assessment of anti-cancer products, the media were centrifuged at ۱۰۰۰۰ rpm for ۱۰ min and the supernatant was separated. Also, the crude extract were extracted from the cell free supernatant with ethyl acetate (۱:۱ V/V). Finally the anticancer effects of the crude extracts and supernatant on MCF-۷ tumor cell were evaluated using the MTT assay method.Results : The isolated actinobacteria were identified by morphological and microscopic criterias. Nine strains were isolated from different samples. They were named EZ (isolated from plant root), Ga۳۱, Ga۵, Ga۲۰, Ga۱۸ (isolated from soil), Mar۱, Man۱, Sa, Red۲ (isolated from Persian Gulf water). The isolates showed anti-cancer activity against the MCF-۷ cell line. The Ga۳۱ strain had the highest anti-cancer (۷۵%-۹۰%) activity against MCF-۷ cell line.Conclusion : The results of this study showed the products of isolated strains have the ability to inhibit tumor cells and these ecosystems can be resources of anti-cancer compounds. The Ga۳۱ strain had the best anti-cancer effect against MCF-۷ cell line. Therefore can be done further research for Purification of these compounds and used the products to in-vivo studies.
