Background: The TP۵۳ gene is the most important tumor suppressor gene inhumans. The aim of our study was to determine the genotype frequency of threecommon TP۵۳ polymorphisms (codon ۷۲ BstUI and intron ۶ MspI, as well as theintron ۳) in a group of Iranian women with and without breast cancer.Methods: Paraffin-embedded specimens of ۶۵ malignant breast cancer casesand ۶۵ cases with benign breast lesions were investigated for the presence of threecommon TP۵۳ polymorphisms by polymerase chain reaction. Samples weregenotyped by polymerase chain reaction followed by variant specific restrictionenzyme digestion.Results: In our study, age grouping as >۵۰ and ≤۵۰ showed that the highestnumber of cancerous and non-cancerous patients was in the age group under ۵۰;according to statistical tests, the difference was significant and recessive alleles ofall three hot spots of TP۵۳ had the highest frequency in the cancerous group. Themajority of the cases with recessive alleles of all three hot spots of TP۵۳ were inthe age group ≤ ۵۰. The difference between cancerous and noncancerous groupswas statistically significant.Conclusions: Our results indicate that recessive alleles in three hot spots ofTP۵۳ gene might play a role in the breast cancer development, especially in womenyounger than ۵۰ years.
