Cytochalasin H cytotoxic effect on human malignant glioma cell line (U۸۷MG) and PLAU and PCDH۱۰ genes expression

Background and Aim: Glioblastoma is the most and common brain tumors malignant. The treatment of glioblastoma is very difficult, due to extensive tissue surrounding of the brain and the aggressive attack. Therefore, the investigation of new therapeutic approaches for this disease is very important. The aim of current study was to provide a proof-of-concept on the mechanism of PLAU and PCDH۱۰ gene expressions after treatment of malignant human glioma cell line (U۸۷MG) with cytochalasin H.Methods: In the present experimental study, we have examined cytochalasin H cytotoxic activities on U۸۷MG cells in vitro for the first time. The cells were cultured and treated with ۱۰-۵ - ۱۰-۹ M of cytochalasin H for ۲۴, ۴۸ and ۷۲ hours. The assessment of cell viability was carried out by MTT assay at ۵۷۸ nm. mRNA expression changes of PLAU and PCDH۱۰ were then evaluated by qRT-PCR.Results: MTT assay showed that cytochalasin H (۱۰-۵ M) inhibited the U۸۷MG cancer cells proliferation after ۴۸ hours. Analysis of qRT-PCR showed that the PLAU expression was significantly decreased in comparison with the control (P<۰.۰۵). The expression of PCDH۱۰ also showed a significant increase when compared to the control (P<۰.۰۰۱).Conclusion: This study shows the effect of Cytochalasin H cytotoxic activities on U۸۷MG cells in a dose- and time-dependent manner and Tumor Regulatory Genes in The Cytotoxic Effect of Cytochalasin H on Malignant Human Glioma Cell Line.
