Background and Aim: Type ۲ diabetes is a disorder of glucose metabolism that results in elevated blood sugar. The etiology of type ۲ diabetes is complex and various genetic and environmental factors are involved. rs۳۵۸۲۹۴۱۹ polymorphism of the NLRP۳ gene Increases IL-۱β production and seems to increase IL-۱β in the development of type ۲ diabetes. The aim of this study was to investigate the possible association of rs۳۵۸۲۹۴۱۹ polymorphism of the NLRP۳ gene with type ۲ diabetes mellitus.Methods: In this case-control study, DNA was extracted from the blood samples of ۱۵۰ individuals with type ۲ diabetes and ۱۵۰ healthy individuals as control samples. SNP rs۳۵۸۲۹۴۱۹ of the NLRP۳ gene was analyzed by Arms-PCR technique and data were analyzed by SPSS software.Results: The results of this study showed that the prevalence of CC, CA, AA genotyping for NLRP۳ gene in the patient group was ۷۰%,۲۹%,۱%, respectively, while in the healthy population it was ۴۱%,۵۷%, and ۲%, respectively (P = ۰.۰۰). The evidence was that In the study population, polymorphism rs۳۵۸۲۹۴۱۹ gene NLRP۳ was significantly associated with type ۲ diabetes. The CC genotype (OR=۳.۴۰۴) and allele C (%۵۴.۹prevalence in the patient's population (P = ۰.۰۰) and OR=۲.۳۸) were considered risk factors for type ۲ diabetes.Conclusion: The polymorphism rs۳۵۸۲۹۴۱۹ of the NLRP۳ gene in code ۲۱۱۳ allele C is converted to A, where the modified protein Q۷۰۵K is formed. This protein is a type of gain of function and ultimately increases IL-۱β production and inflammation and thus plays a role in pathogenesis. There has been no study showing the association of polymorphism rs۳۵۸۲۹۴۱۹ gene NLRP۳ with type ۲ diabetes. However, in studies conducted by various researchers, polymorphism rs۳۵۸۲۹۴۱۹ gene NLRP۳ in the pathogenesis of rheumatoid arthritis, celiac disease (allele A was identified as a risk factor) And MS is involved in Crohn's disease (allele C was identified as a risk factor). The results of the present study showed the association of polymorphism rs۳۵۸۲۹۴۱۹ gene NLRP۳ with type ۲ diabetes (allele C was identified as a risk factor). The reason for the discrepancy between risk factors in higher studies could be the difference in populations, diseases, different races and intraction between genes and SNPs, which means that allele C may be present in the presence of various factors and other genes and SNPs. It has found a part and is considered as a risk factor for type ۲ diabetes.
