Crossbreeding effect on GDF۹ and FecB gene SNPs and their protein structure

چکیده مقاله:

Background and Aims: SNPs are considered as an important genetic marker that is considered as a vital genetic source responsible for phenotypic changes within the species. Reproduction is one of the most important economic traits of sheep.Methods: The rate of twinning and multiplication in Arabi sheep is low. so to improve this trait, crossbreeding with Romanov rams was done. For this purpose, ۱۰۰ Arab ewes were registered, of which ۹۰% with Romanov ram and ۱۰% with Pakistani ram were laparoscopically inseminated (intrauterine). This continued until ۳rd generation (F۳) was acquired. In the next step, blood samples were taken from ۵۰ animal (F۳ population) and its DNA was take out by salt extraction method. Then, the extracted DNA was amplified by PCR and the PCR product was sent to MicroSync Switzerland for sequencing. After receiving the sequences, they were read using BIO EDIT software and compared with the pattern sequences by NCBI BLAST.Results: An SNP was observed in the intron of the GDF۹ gene and no SNP was found in the FecB gene.Conclusion: Since SNP was not found in one gene and in the other gene, found in its intron, no effect on the protein structure of these two genes could be imagined.
