Background and Aim: The mitochondrial DNA (mtDNA) involves the control of cell activities such as ATP production, heat production, cell differentiation, cellular respiration, lipid metabolism, steroid hormone synthesis, regulation of apoptosis, and calcium storage. A total of ۱۳ proteins encoded by mtDNA form components of the respiratory chain or oxidative phosphorylation system and are critical for the production of ATP. Many studies in human have reported that mtDNA variation resulted in metabolic and developmental disorders. In livestock, association of mtDNA polymorphisms with growth, reproduction and production traits were reported. The aim of this study was to investigate the polymorphisms of the mtDNA ATPase۶ gene in Snajabi sheep breed.Methods: . For this purpose, blood samples were collected from ۱۰۰ ewes and DNA extraction was carried out by salting out method. The polymerase chain reaction (PCR) was then performed using a primer pair (F: ۵′-ATTCTCCTTGATGATATGCCAC-۳; R: ۵′-CTGGCCTCCAATTCATGTGAG-۳′) to amplify a ۸۹۵ bp fragment of mtDNA that contained ATPase ۶ gene. To identify polymorphisms in this region, the SSCP technique using ۶% polyacrylamide gel was performed.Results: . The results showed that six different patterns (A, B, C, D, E, and F) was observed in this breed. The A pattern had the highest frequency of ۴۸%, and the C and E patterns had the lowest frequency of ۹%. Furthermore, the B and D patterns had modest frequency with respect ۱۴% and ۲۰% frequencies.Conclusion: The results of this study confirmed the polymorphisms in this gene in the Sanjabi sheep breed.
