Effect of β-Cyclodextrin Nanocavity Complexation on Solubility, Stability and Antioxidant Activity of Gabapentin

Gabapentin(GBP) was initially development for the treatment of pain, but is currently used for various uses, especially pain with a neurological origin. β-Cyclodextrin (β-CD) on the other hand due to its property of encapsulating molecules that are hydrophobic in nature is widely applied as drug delivery vehicle. Gabapentin within β-CD complexes were evaluated by dissolution studies, Fourier transform infrared (FTIR) and Ultraviolet-Visible (UV-Vis) spectroscopic methods. Phase solubility studies showed an enhanced solubility of GBP with increasing amount of βCD. The stoichiometry of this inclusion complex was established to be equimolar (1:1) and its equilibrium constant was calculated byusing a modified Benesi-Hildebrand equation at room temperature. Antioxidant activity studies on complexes carried out with DPPH tests. Analyses results showed that the formation of the complexes resulted in enhanced solubility with increased its antioxidant activity of gabapentin. The prepared inclusion complexes represent potential pharmaceutical active substances for a new products design.
